用酶细胞化学方法结合超微结构,观察了家兔输精管经金属环夹及加不同性质支撑物后输精管碱性磷酸酶(AKP),睾丸乳酸脱氢酶(LDH)的活性变化及输精管、睾丸和附睾的超微结构改变,在细胞学基础上,探讨了输精管金属环可复性节育术的影响因素,并结合酶活性变化,分析了其复通后复孕率较低的原因。
Abstract:Objectives:To study the changes of alkalime phosphatase(AKP),lactate dehydrogenase(LDH)activi- ties and ultra- structures of vas deferens ,testis and epididymis of rabbit after occlusion with ring clips.Design:Experimental animal study.Setting :Laboratory in Jinan Military Hospital.Animals: 24 male normal mature rabbits of2.5~3.0kg in weight,with sperm count over 160 million/ml and without antisperm antibodie:V- shaped titanium alloy clips were placed on vas deferens supported with a piece of No.1silicon rubber nylon thread(A) ,or with 180~200 μm horse tail hair(B),or with no support(C). Theanimals were divided into 3 groups with 6 in each group.Semen andlysis was performed 2 weeks post opera-tion and repeated once every 2~3weeks until removal of the supporting material. Specimens of vas deferenswere prepared for AKP and electron microscopy(EM )examinations.Testis specimens were prepared forLDH and EM examinations.Results:Reduction of AKP in the animal groups with ring clips,more marked in the B group,than thecontrol group. Reduction of LDH in the treated groups was seen mainly in the Sertoli cells, spermatozoa andsperms. EM examination showed edema of vas epithelial cells and irregular arrangement of microvilli,de-crease of spermatozoa,slight atrophy of,eminiferous,tubules and disturbance of spermatogenesis.Thechanges were more marked in the groups with clip and support.Conclusion:The results indicated the ring clips and supporting material caused damage of vas epithelialcells and testis seminiferous tubules and hence the disturbance of enzyme activities and spermatogenesis.
